Alessandro Matri was born in Sant’Angelo Lodigiano, Italy. Local Serie D club A.C. Fanfulla 1874 was the first club in young striker’s career. Alessandro was scouted by A.C. Milan back in 1996 and eventually signed a long-term contract with the Rossoneri. For 6 years Matri played for Milan youth teams before ascending to the ‘Primavera’ Milan squad.
In May 2003 Alessandro Matri made his debut in Serie A against Piacenza, replacing Roberto Bortolotto in the 71st minute of the match. During 2004/05 season Matri played on loan at A.C. Prato and scored 11 goals in 32 appearances. For the following 2005/06 season he was sent out on loan again, this time to A.C. Lumezzane, to net 13 goals in 32 matches. In 2006/07 season Matri continued his loan streak at A.C. Rimini side, featuring in 28 games and scoring 4 goals.
In 2007 Alessandro signed a permanent contract with Cagliari Calcio. The club bought half of the rights for the player from Milan for €2.3 million. Matri made his debut at the new club in a match against Napoli and scored his first goal for Cagliari against Parma.
During 2007/08 season Matri made 34 appearances, scoring 6 times. Despite a large number of appearances Alessandro was a regular substitute because of Robert Acquafresca’s more impressive performance. During the following season Matri played 31 match and only 11 of them in the starting lineup.
In 2009/10 season, Acquafresca left the club and was replaced by Matri. Alessandro scored 13 goals and became Cagliari’s best goleador. In the same season he broke Luigi Riva’s record by scoring goals in 7 matches in a row. In 2011 Juventus and Cagliari agreed a loan deal for Matri worth €2.5 million. After a loan spell Matri signed a permanent deal with The Old Lady and fired 29 goals in 84 matches during two seasons.
In 2013 the forward returned to AC Milan but was not able to break into the main squad and was later loaned to Fiorentina.
